Dr. Liu's research group is interested in studying the expression and functions of galectin-3, -7 and -12, in particular the roles of these proteins in inflammation and neoplasm.
Galectin-3 is a pleitropic protein expressed in several leukocytes in a regulated manner. In particular, this lectin is present in mast cells, macrophages and dendritic cells, and present in subsets of lymphocytes when activated. Mast cell mediator release and dendritic cell motility are dysregulated in the absence of galectin-3, and involve atypical signaling through cell surface receptors in both cell types. Limited analyses of several cell surface receptor levels in gal3+/+ and gal3-/- cells analyzed by flow cytometry reveal similar densities. However, in light of galectin-3 involvement in cell surface glycoprotein trafficking in epithelial cells, it is possible that this lectin has similar function in leukocytes and regulates glycoprotein sorting or glycan densities of membrane receptors. As cell surface glycans participate in signaling events of membrane receptors and in intercellular communication, knowledge of glycan composition and densities on leukocytes would contribute to our understanding of these structures in cell communication. We will generate dendritic and mast cells from bone marrow cultures, and lymphocyte populations from spleen by magnetic selection, of gal3+/+ and gal3-/- tissues. Duplicate samples of the following gal3+/+ and gal3-/- cells will be provided to the Consortium for glycan analyses.
1. mast cells sensitized +/-IgE
2. dendritic cells activated +/-LPS
3. CD4 T cells activated +/-CD3/CD28
4. B cells activated +/-LPS
