We study the role of glycosylation in ion channel function. Specfically, we are focusing on how ion channel glycosylation modulates, controls, and impacts cardiac, skeletal muscle, and neuronal electrical activity.

Experiment to be conducted:
We are interested in understanding the role of differential glycosylation in modulating excitability throughout the developing myocardium. We have recently shown that differential sialylation of the cardiac voltage-gated sodium channel alters channel function throughout the developing myocardium. In addition, through the Consortium, GeneChip analyses of neonatal and adult mouse atria and ventricles have indicated significant differential expression of glycosyltransferases/glycosidases. We now wish to extend these GeneChip and functional (ion channel) studies to determine differences among the glycan structures throughout the developing myocardium. As such, we propose to utilize the power of Core C to determine the glycan structures expressed on atrial and ventricular myocytes of newborn and adult mice. Such data could be correlated with the GeneChip data, and serve as significant support for our sodium channel functional data. In addition, the correlation of such Core C and Core E studies would help meet a priority of the Consortium. We could provide the tissue samples needed. Our GeneChip anlayse
s were done using mixed gender triplicate samples for each myocyte type (atria/ventricle,newborn/adult). I would suggest a similar strategy for the glycan profiling studies. Details of the amount and types of samples needed could be determined at a later date in consult with Stuart Haslam.

GBP being Addressed:
Cardiac voltage-gated ion channels; cardiomyocytes
Specific aims being addressed:
* 2. Establish the cell types involved in communication
* 4. Determine how GBP-ligand interactions mediate cell communication
* 6. Identify the glycosyltransferases that synthesize carbohydrate ligand(s)
* 7. Determine whether regulation of glycosylation modulates GBP function
